Responses of Populus spp. to Zn excess: carbon assimilation, structural modifications, metal distribution and cellular localization, identification of target genes

Riassunto: Poplar is a multipurpose tree species, is promising for energy, paper and pulp production, for growing in contaminated soils, and as model system in tree physiological and biomolecular studies. In previous studies we have reported about physiological and biochemical responses of hybrid poplar clones to several heavy metals (Zn, Cd, Cu, etc.), in different growing conditions. These researches are part of a broader project, which includes in vitro, growth chamber, glasshouse, and field experiments aimed to test the tolerance of poplar genotypes to heavy metals. Using clone I-214 as a model system for poplar physiology and molecular biology, we identified 1 mM Zn as the crucial dose influencing plant biomass, photosynthetic parameters, glutathione metabolism, structural traits and metal localization/distribution in leaves and roots. To analyse transcriptomic changes in Zn-exposed polar leaves, we used microarray technology by Affymetrix GeneChip Poplar Genome Array, containing more than 61,000 probe sets (over 56,000 transcripts and gene predictions). In a set experiment in which the 1 mM Zn treatment was compared to the basal Zn supply in nutrient solution, 3,860 sequences resulted differently expressed. We analysed the correspondent ESTs classified into three different significance levels (P < 0.01; 0.01 < P < 0.03; P < 0.05) and we tried to identify specific genes involved in I-214 (and Populus) tolerance mechanisms to Zn and other metals. The final aim of the present study was to clone the full length sequences, to serve as basis for the up coming annotation of the Populus genome sequence in the frame of further studies of poplar response to metal stress.
