Log traceability and supply-chain verification by molecular markers in Fagus sylvatica wood in Italy

Riassunto: Matching a timber log into its population of origin would signify an important improvement in the context of log traceability and in the preservation and valorization of wood chain. Spectrometry and isotopic methods have been applied and proposed to differentiate wood samples from different geographical origins. However, these approaches are influenced by the local environment and variability of chemical composition that in some cases can bring to show a discrepancy between individuals from the same population or even between different tissues from the same individual. For this reason, major advances can be obtained in the application of DNA analyses on timber, to support the determination of identity and provenance. Chloroplastic DNA (cpDNA) markers are able to show the geographical structure of populations and could be used to differentiate timber’s origins from one another. Additionally, highly polymorphic nuclear microsatellite (nuSSR) markers could be used to generate DNA profiling databases for individual/species identification. Both of these tools require the development of large comprehensive databases. cpSSR and nuSSRs have been applied to wood material of beech (Fagus sylvatica) coming from four “reference” cutting sites and putatively same wood material collected in sawmills in Italy. The four source origins were clearly identifiable. The individual assignment test by the Bayesian multilocus-approach correctly self-assigned “unknown” individuals in 78% of the samples, with higher uncertainty in one site, where further samples are being processed. These results represent a first attempt to delineate a match between wood reference material of beech from different areas in Italy and unknown material collected at the sawmill. The application of molecular markers could act as a benchmark, for further development of an independent, scientific, supply-chain verification, that should be implemented into the inventory process.
